Regulation of GTPase Activity

The involvement of G-protein signaling in a wide variety of cellular activities has resulted in the study GTPases and their regulators as therapeutic targets in a number of human diseases. The regulation of GTPase signaling is highly and carefully regulated and hundreds of GTPase regulators have been identified.

OxiSelect Protein Carbonyl Assay Kits

Cell Biolabs OxiSelectTM Protein Carbonyl Assay Kits
The most common products of protein oxidation in biological samples are the carbonyl derivatives of Pro, Arg, Lys and Thr residues. Such derivatives are chemically stable and serve as markers for oxidative stress in most types of reactive oxygen species.

Cell Biolabs’ OxiSelect™ Protein Carbonyl Assay Kits provide rapid, efficient methods for detection of protein carbonyls. Three assay formats are available: immunoblot, ELISA or spectrophotometer. All formats are suitable for use with purified protein, plasma, serum, or cell lysate samples.

Protein Carbonyl ELISA Kit
• Sensitive: Detects samples as low as 10 μg/ml
• Greater Sample Retention: No concentration/TCA precipitation steps

Protein Carbonyl Immunoblot Kit
• No Molecular Weight Shift: DNPH Derivatization after immunoblotting allows direct comparison of oxidized and non-oxidized protein fingerprint

mTOR Pathway Antibodies

Studies of mTOR (mammalian target of rapamycin) have demonstrated that it performs an essential role in integrating cellular signals received from growth factors and from the detection of nutrient, stress, and energy levels in the cell.
The integration of signals by mTOR functions to control cell growth by influencing cellular processes such as translation initiation, ribosome biogenesis, and transcription factor localization.

Bethyl have a range of Polyclonal Antibodies to Proteins in the mTOR Pathway.

PhosphoSeek™ Assays

We are pleased to offer the new PhosphoSeek™ assays from our supplier BioVision. These assays are based on a novel fluorescence technology that does not require antibodies or radioactive labels and detect enzyme activity and inhibition of:

1. Phosphodiesterase 5A
2. Phosphoinositide 3-kinase
3. Sphingosine kinase
4. Protein tyrosine phosphatase 1B

PhosphoSeek™ assays utilise a proprietary small molecule sensor containing a trivalent metal ion which binds to phosphoryl groups on biological substrates.

KinaseSTAR JNK Kits

The JNK Activity Screening Kit is used to screen JNK activity relatively quickly for large numbers of samples. The assay utilizes a N-terminal c-Jun fusion protein bead to selectively pull down JNK from cell lysate. c-Jun phosphorylation is then measured using a phospho-c-Jun specific antibody.

The KinaseSTAR JNK Activity Assay Kit utilizes a JNK-specific antibody to immunoprecipitate JNK from cell lysate. JNK-specific activity is then analyzed by Western blot analysis of the phosphorylation levels of c-Jun using a phospho-c-Jun specific antibody. The kit specifically detects JNK activity, other kinase activities would not be detected.

Biological Application of FRET – Protease Detection

In recent years, FRET based assays have found broad applications, one of which is the detection of protease activity. Proteases play key roles in protein activation, cell regulation and signaling, as well as in the generation of amino acids for protein synthesis or utilization in metabolic pathways.

FRET-based assays have made the detection of protease activity and High Throughput Screening of protease inhibitors faster and easier. AnaSpec, a world leader in FRET peptide technology, is proud to be the first in the industry to offer long wavelength protease assays for:

Cathepsins D, S
HCV (Hepatitis C Virus)
HIV
MMPs
α- (TACE), ß-Secretase (BACE1)
Renin
and WNV (West Nile Virus)
