What are PARP proteins?
Poly(ADP-ribose) polymerase (PARP) proteins are essential for DNA repair within cells, repairing single-strand DNA breaks through the base excision repair pathway. Inhibiting PARP proteins has become a promising approach in cancer therapy, particularly for tumours with faulty DNA repair mechanisms like those with BRCA mutations. PARP inhibition hampers the repair of single-strand breaks, leading to the build-up of double-strand breaks during DNA replication. In cancer cells lacking efficient homologous recombination, these double-strand breaks are especially harmful. This weakness is exploited by PARP inhibitors within drug discovery, which selectively target cancer cells with defective DNA repair.
