► Preloaded LNPs
Cayman's preformulated LNPs containing reporter mRNA are intended for proof-of-concept experiments to help determine whether SM-102-based LNPs can effectively lead to expression of proteins of interest in target cell types (in vitro or in vivo).
Transfection of cells with mCherry-encapsulating SM-102 particles: Huh7 hepatocytes (left) and A549 lung epithelial cells (right) were incubated with LipidLaunch SM-102 (mCherry) at an RNA concentration of 150 ng/ml for 24 hours in cell culture media with 10% FBS. Cells were stained with 4 uM Hoechst 33342 and imaged using 590 LED/Texas Red™ and 365 LED/DAPI cubes on an imaging plate reader.

Cayman's LipidLaunch Loadable LNP SM-102 Kit provides cargo-ready, lyophilised LNPs while designed to facilitate easy and efficient RNA transfection with reduced toxicity when compared to traditional methods.
Loadable LNPs protocol summary: Following rehydration, loadable LNPs are mixed with RNA cargo before the addition of cell-based assay buffer to promote encapsulation. The loaded LNPs are then ready for downstream delivery to target cells.

Cayman’s LipidLaunch LNP-102 Exploration Kit provides a starting point for researchers to explore the feasibility of using SM-102-based LNPs for specific applications without any requirement for specialised equipment.
Firefly luciferase mRNA encapsulated in LNP-102, LNP-0315 and LNP-MC3 injected subcutaneously into CD-1 IGS mice: In vivo luminescence with intraperitoneal injection of D-luciferin) captured by IVIS 8 hours later.
► LNP Uptake Kits
LNP uptake kits enable evaluation of LNP uptake into your cells of choice, using sterol-based fluorescent tracers. Also compatible with other biological readouts.
LNP-102 uptake in hepatocytes: LNP-102 was formulated using a FRR of 3 and a TFR of 15 ml/min. After dialysis against PBS, LNPs were diluted 1:200 in cell culture media and added to Huh7 hepatocytes for 48 hours. Imaging was carried out in PBS on an imaging plate reader at 20X with a GFP LED/filter cube.
