An inhibitor of ATM kinase (IC50 = 0.78 nM); selective for ATM over ATR, PI3K, mTOR, and DNA-PK (IC50s = >30, >12, >16, and >29.9 µM, respectively); sensitizes glioma cells expressing mutant, but not wild-type, p53 to ionizing radiation; reduces the growth of brain metastases and increases survival in an NCI H2228 mouse xenograft model at 20 mg/kg twice daily, p.o., in combination with radiation
